Preparation and characterization of selenium nanoparticles modified by Tremella polysaccharides and the anti-canine mammary tumor activity.

Abstract
In this study, a novel type of selenium nanoparticle (SeNPs-TP) was synthesized by incorporating Tremella polysaccharide (TP) during the chemical reduction of sodium selenite/ascorbic acid. A minimum particle size of SeNPs-TP, 62.7 ± 8.21 nm, was obtained under optimized single-factor conditions. The synthesized SeNPs-TP were characterized using by dynamic light scattering (DLS), scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS), ultraviolet-visible (UV-Vis) spectroscopy, and Fourier-transform infrared (FTIR) spectroscopy. The results demonstrated that SeNPs-TP markedly enhanced the stability of SeNPs. In vitro cell assays showed that SeNPs-TP exhibited stronger anti-proliferative and anti-migratory effects on canine mammary tumor cells (CMT-7364) than 5-FU, while demonstrating better safety toward normal canine mammary epithelial cells (CMEC-828). Additionally, further analyses indicated that SeNPs-TP induced apoptosis via the mitochondrial pathway. In a mouse xenograft tumor model, it was found that SeNPs-TP significantly inhibited tumor growth and metastasis without observable toxicity to major organs. These findings suggest that SeNPs-TP may serve as a promising chemotherapeutic candidate for the treatment of canine mammary tumors (CMT).
